Effect of medium osmolarity on hybridoma growth, metabolism, and antibody production.

چکیده
Osmolarity is an important process variable during the cultivation of mammalian cells in vitro. Cell culture medium is designed to have osmolarity in the range of 260 and 320 milliosmoles (mOsm), basically to mimic the osmolarity of serum at 290 mOsm/kg. Fragmented information on the response of hybridoma cells to elevated osmolarity is available. A decrease in cell growth and an increase in antibody production were reported at high o~molarit ies.~~ Influence of osmolarity on antibody productivity seems to be cell line dependent.12 Here we report the effects of osmolarity on hybridoma cell growth, metabolism, and antibody synthesis. The osmolarity of the medium was altered by the addition of both ionic and non-ionic substances.

Effects of ammonia and lactate on hybridoma growth, metabolism, and antibody production.
The influence of ammonia and lactate on cell growth, metabolic, and antibody production rates was investigated for murine hybridoma cell line 163.4G5.3 during batch culture. The specific growth rate was reduced by one-half in the presence of an initial ammonia concentration of 4 mM. Increasing ammonia levels accelerated glucose and glutamine consumption, decreased ammonia yield from glutamine, ...
متن کاملEffect of initial cell density on hybridoma growth, metabolism, and monoclonal antibody production.
A murine hybridoma cell line (167.4G5.3) was cultivated in batch mode with varying inoculum cell densities using IMDM media of varying fetal bovine serum concentrations. It was observed that maximum cell concentrations as well as the amount of monoclonal antibody attainable in batch mode were dependent on the inoculum size. Specifically, cultures with lower inoculum size resulted in lower cell ...
متن کاملEffective antibody production by reusing culture medium previously used in antibody purification.
Culture medium used in antibody purification contains growth factors, but is usually discarded after antibody purification. We examined to determine whether the growth factors can be (i) recovered and (ii) used in subsequent cell culture. Using this medium enhanced the survival of hybridoma cells and improved antibody production. Culture medium previously used in antibody purification offers an...
متن کاملEffects of Environmental Conditions on High-Yield Magnetosome Production by Magnetospirillum gryphiswaldense MSR-1
Background: Magnetotactic bacteria are a heterogeneous group of Gram-negative prokaryote cells that produce linear chains of magnetic particles called magnetosomes, intracellular organelles composed of magnetic iron particles. Many important applications have been defined for magnetic nanoparticles in biotechnology, such as cell separation applications and acting as carriers of enzymes, antib...
متن کاملProduction of monoclonal antibody using free-suspended and immobilized hybridoma cells: Effect of serum.
The effect of serum on cell growth and monoclonal antibody (MAb) productivity was studied in a repeated fedbatch mode using both free-suspended and immobilized S3H5/gamma2bA2 hybridoma cells. In the suspension culture, serum influenced the cell growth rate but not the specific MAb productivity. The average specific growth rate of the suspension culture in medium containing 10% serum was approxi...
